Bloodstream infections (BSIs) are triggered by the existence of pathogens in blood and considered a major health burden worldwide, especially when they result sepsis septic shock. Common diagnostic methods time-consuming, present low specificity, or suffer from interference components, which hampers timely effective treatment BSIs. In this work, novel microfluidic assay was developed combining bead-based chip bacteriophage receptor binding proteins (RBPs) as extremely specific sensitive recognition molecules for multiplex concentration detection Escherichia coli Pseudomonas aeruginosa , highly prevalent bacteria The device comprises microcolumn antibody-functionalized agarose beads were packed allowing entrapment target bacterium blood, providing its separation. Doripenem, a parenteral carbapenem, exhibited high affinity for penicillin-binding protein 2 (PBP2) and PBP3 in Pseudomonas aeruginosa and PBP2 in Escherichia coli, the primary PBPs whose inhibition leads to cell death.
